I was playing earlier with Strong as a companion and almost fell off the couch laughing. Some random ghoul came creeping out of a house and Strong whacks it with a baseball bat and mumbles "filthy ghoul". I don't know why but it was probably the funniest thing I've witnessed playing this game so far.

I'd say the main character's lines when you skip dialogue. My favorite is when you call Sturgis a stupid grease jockey (or grease monkey maybe...can't remember which). Or, when you're talking to mama Murphy and just go, ugh...old people.

You have to press the right arrow button to get those kinds of responses.
My favourite ones were "bimbo" and "bitch" when talking to Tina De Luca.

I was talking to ada after her caravan just got smashed and all friends killed, while in the dialogue, a random scavenger walks up and says 'Hmm there's some nice stuff here' to himself. Turns out he was looting all the stuff from the caravan.

A raider told me a dad joke. Yelled out "Now things are heating up" and threw a molitov at me.

Very early in the game, before I realized that a companion will arrive anywhere you go I thought I needed to make sure Dog Meat got on the elevator with me at Corvega. The door closed on him, his head sticking through the door and when the elevator went up his head went, to my point of view down. Horrified I went back down and he emerged up through the floor, cracked me up.

I was going up the elevator to fizztop one day and MacReady was in front of me not-quite-on the elevator when he started having his 'never get tired of hearing me say we're best friends' talk with me and WHAM he is hit on the top of the head by the edge and is more or less squashed and falls down off the elevator and next thing I hear is him yelling about his leg and 'heal your companion' comes up.' I died laughing.

Nora and Piper walk into the room in a vault where there had, a long time ago, been AA meetings. A bunch of folding chairs in a circle and some skeletons on a few of those chairs.
And Piper says something about musical chairs. I can't remember exactly what... meant to write it down but was laughing too hard

The deep soul searching conversation between two Super Mutants, who were discussing their same-mess...they ended up threatening to eat each other. It was hysterically funny.
The funniest line in Fallout 4 is from NW's World of Refreshments. Something about Apple Juice not being sold today because it got mixed up with Lion Urine.

Bethesda games can provide some of the best/weirdest/stupidest timing or oddities that produce comedy gold. The first one from FO4 that comes to mind wasn't even that unique, just a fantastic physics glitch.
I tossed a grenade 30 yards or so at a raider perched behind a fence, he shot 50 feet straight up in the air. Ragdoll physics bugs ftw.
